ATAC-Seq Kit常见问题
1、此试剂是否和Buenrostro et al在nature上发表的方法一样？
Active motif 基于最初Buenrostro 的文章和2017年Corces et al. 在nature上发表的文章，优化了缓冲液以满足跟多的实验需求和高效的实验过程。
2、试剂盒中包含的重组Tn5酶的来源是？
此试剂盒中的Tn5是由我司克隆，表达，纯化，验证的超活性突变克隆。所提供的这款转座酶预先已链接好用于NGS测序的接头（adaptor）序列，所以它不许任何预处理过程，可直接用于实验。
3、每个反应需要使用的小的细胞数量或者组织量是多少？
50,000个细胞或20 mg组织样本。
4、可以使用冻存的组织和细胞吗？
可以，但样本得保证高质量且高活性，细胞应在控制速率冷冻中冷冻保存（例如细胞冻存液梯度冻存），冷冻介质的配方应防止冰晶形成和随后的细胞损伤，组织应在-80°C下快速冷冻。
5、可将细胞核冻存后用于后续ATAC-seq实验吗？
可以的，提取的细胞核可以-80℃冻存用于后续使用。制备的同样品细胞核可合并在一起增加样本量。
6、可以使用FACS分选后的细胞吗？
可能可以。但FACS后细胞可能会被破坏。一个高质量的ATAC-seq 文库需要高质量高活性的细胞。
7、您是否建议使用40微米筛孔的细胞样本过滤器？
是的，我们推荐使用Falcon 40微米过滤器（Falcon货号：352340）。
8、所有样本都需要DNase处理吗？
DNase的处理能够帮助确定悬液中游离DNA的去除，如线粒体DNA的去除，如果细胞样本中有15%的死亡率，细胞活性不够且必须使用此酶，它最终可能会移除你想要的细胞数据。只有当细胞是活较高时，并且不会被酶消化时，才建议将其作为一种选择。
9、若没有thermomixer，是否还能使用此试剂盒？
建议使用Thermomixer。然而，我们已经对此进行了测试，在37°C温度下不使用thermomixer而是在带热盖的热循环器中进行转座反应，反应能够成功。
10、 是否能够室温融化此试剂盒包含的所有试剂盒组分，如master mixes和酶？
除了转座酶和Q5 DNA polymerase 以外，其他试剂均可室温融冻。转座酶和Q5 DNA polymerase 包装中含有甘油，不会被冻结，请直接放在冰上使用。其他组分可融冻多次用于以后使用，所以不需要一次性将16次的试剂用完。
11、 每个样本需要测序多少量的reads？
常规20 M reads的测序量就足够了。
